Background :
@EN Self-regulating insulin delivery system using glycosylated insulin has been developed as one of the mthods controlling blood glucose levels more physiologically. In this system, as blood glucose level rises, glucose is replaced with the
glycosylated
insulin at its binding site of lectin. Succinyl amidophenyl-a-D-glucopyranoside insulin (SAPG-insulin) is one of the glycosylated insulin widely used in self-regulating insulin delivery system. Although SAPG-insulin has been reported to reduce
blood
glucose in rats, bioactivity in human has not tested.
@ES Method :
@EN Seven NIDDM patients treated with diet and exercise were studied. SAPG-insulin was synthesized by the method of Jeong et al, separated by con A affinity chromatography. After on overnight fast, 0.1 U/§¸ of either SAPG-insulin or Velosulin(r)
were
injected, in random order, subcutaneously into the abdominal wall. Venous blood samples were obtained to determine plasma glucose, insulin and C-peptide concentrations.
@ES Results :
@EN Basal plasma glucose levels before subcutaneous injection of SAPG-insulin were not different from those before Velosulin(r) injection. Decrements of plasma glucose and C-peptide levels and the time to nadirs were not different. Increments of
plasma
insulin levels after injection of SAPG-insulin were larger than those after injection of Velosulin(r). Side effects of SAPG-insulin such as fever, skin rash and itching sensation were not found.
@ES Conclusion :
@EN The hypoglycemic effect of subcutaneously injected SAPG-insulin was not different from that of Velosulin(r). No significant side effects of SAPG-insulin were noted. These results suggest that SAPG-insulin can be an effective agent employed in
developing chemical self-regulating insulin delivery system.
